Abstract
A simple but efficient continuous-flow polymerase chain reaction (CF-PCR) system was newly developed. It has successfully amplified a human genome DNA in less than 10 minutes using 30 cycles' PCR. A unique design of a small polymer disk, equipped with a long spiral microfluidic channel of varied width, was utilized. The disk was fabricated with polymer replication and thermal bonding techniques, which is suitable for mass production and single use. For a stable and fast PCR, uniform temperature and fast thermal response were realized by sandwiching the disk with metal heat blocks of constant temperature. The simple fluidic and thermal control is suitable for miniaturization. The unique architecture of the disposable polymer disk for CF-PCR system produced enhanced PCR performance in speed, size, and control.
